Is possible to have AI planes fly a specific route between two airports.For example, take-off, turn left to 270 degrees fly direct to Yoo NDB then direct destination.

Yes, if you create fake airports in AFCAD at each intersection (elevation = cruise altitude) and code the flight for a stop of less than 5 minutes at that fake airport.
