I've a doubt about tileproxy's use :whenever I start my flight simulator must I start so tyileproxy? Isn't enough tile's download I've made the day before in another fly in the same country?

Yes, start tileproxy first and then FSX.

Tileproxy does need to be running to supply the tiles to FSX. It will use the tiles it needs that have already been downloaded and saved to the cache.
