I have the Windows 10 latest update just install last week and have no problems starting up P3DV4.3. At startup, all of your modules in the dll.xml are loaded along with some of the scenery you already had enabled.

When loading from the setup screen to the scenario start, it will usually hang at 6% if you have a lot of photoscenery enabled.  It is all loading.  Just takes a lot of time.  Orbx scenery will take some time loading too.  From 0% to about 80%, the sim is loading terrain and, if it stopped at 25% some scenery is taking a long time to load.

I have no idea what scenery you have installed.  It is obviously loading the scenery required for the start of your flight.  If it is addon scenery, then I would disable that scenery and try again.  See if it loads faster.  If you are using the Lorby-Si Addon Organizer, you can disable the addon scenery there.  I would also click on the Tools tab and then click on check encoding and make sure all of your P3D config files and xml files are properly encoded.  If not, just click save on the scenery tab and that should encode everything properly.

If you think it was Windows 10, then you need to go to the AVSIM Windows 10 Forum and post your question there or look at the comments from others who might have upgraded to the latest update (released last Tues).  Make sure you start up P3D every time with Admin Privileges for ALL users.  One thing you can do is try using the Restore Point option and, if that does not work, then you may have to reinstall P3D and all of your addons.  Perhaps, if you find what is loading at 25%, you can disable that scenery and find out whether that scenery needs to be reinstalled.  Windows 10 might have changed your file structure or the link to the scenery is no longer valid.  I know I just reinstalled Windows and it changed all of my drives and my addon.xml's were no longer directed to the scenery.
