FSX-MS Is there a way to reload FSUIPC during flight?

My question is, if there is actually a way to reload FSUIPC during flight?

Enrico,

No.. the .dll is loaded when the sim loads.
You can however, re-initialize simconnect if the data stream between the 2 happens to stop or simconnect fails.
It is stated in the doc - "FSUIPC4 for Advanced Users.pdf" around page 33 of 63.

It should also be listed as such in the UI for assignments. 

If you are doing manual .ini edits and want to reload the new edits while the sim is running, then from the UI :
For keyboard keys, in the "Key Presses" tab, click - Reload all Keys
For joystick buttons ( or other hardware ), in the "Buttons + Switches" tab, click -  Reload all butttons
To reload everything, in the "Axis Assignment" tab, click - Reload all assignments
