Question for you, what is the best prop RPM, best throttle setting or inches of manifold pressure, and what is the best altitude above MSL for the DC-6 for the lowest gasoline consumption as possible? Thanks!

Have a look at the manuals. There are power charts near the back that explain everything going into making this determination.

Welcome. Sorry for the vague answer, but there's a lot that goes into fuel consumption: air temp, weight, power setting, pressure, etc. (just about all of which are covered by the charts). I generally get the best burn around 8000, with a lower cruise power setting. Keep in mind, though, that the lower cruise power settings - while they give better fuel flow - mean you're spending more time in the air, burning potentially more fuel.

I read from one of the Alaska-based pilots we had around during testing that the cargo haulers spend most their time at 800BHP cruise below 10,000. They are usually more interested in cost than time, the fuel cost is only part of it. The overhaul on these engines is very expensive and they keep the wear and tear down to a minimum.

 

Pilots used to turbojets expect that higher altitudes yield lower costs, that is true for turbojets but not these beauties.  You get a few higher TAS numbers but at a cost.
